Without question, my answer is Harry S. Truman. The Cooper/Harry "bromance" is one of my favorites in all of TV/movies. I thought Robert Forster did a very good job in his role, but it just didn't feel "right" to me without Harry. Oh well, I understand Michael Ontkean left acting behind to live a quiet life and I fully respect that. I just missed him, is all.
Before anyone asks, I always felt that Annie and John Justice Wheeler were a betrayal of Lynch and Frost's original artistic vision, so I was kind of offended that they existed at all.
As to why Cooper didn't ask about Annie, why didn't he ask about Audrey or any number of characters? Either he did and we weren't shown it, or he just didn't have the time to slow down and focus on anything other than his quest.
Also, he only knew her for like a week. Who cares?
I was never a fan of Annie either - a dull character who was shoehorned into the final stretch of Season 2. I thought it was kind of funny that her character was pretty much written out of the story in The Return. Diane made for a much better romantic interest for Cooper in The Return, even if their relationship was kind of retconned into the story.

As an answer to the question, yes, Harry Truman. It's the one character whose absence was sorely felt, especially as Hawk, Andy and Lucy were all there.
But if Harry had been the sheriff in full capacity, the series might have only lasted 4 episodes, as he would have connected the dots in like 5 minutes after Hawk received the phone call from Lanterman.
Like a lot of people, I'd have loved both Bowie and Ontkean in The Return, although their characters technically did feature (Harry Truman as an offscreen presence and Philip Jeffries in a new form). I'm not sure I can count The Man From Another Place/The Arm either, as we got the Evolution...
Of the characters who didn't feature in The Return (in some way) at all, Chet Desmond is probably top of my list. I mean, where exactly is he??
Annie simply because I've had a crush on Heather Graham since I was 12.
Sam Stanley because I would love to see a scene or scenes with him and Albert together. Lots of comic potential.
