Maybe Mike is communicating with Mrs. Houseman and her New York friends at the Great Northern - thus causing the hum
I think it definitely goes back to the "Mrs. Houseman" conversation in episode 1. There's just no way that was the first dialogue in the Return that takes place in the town of Twin Peaks for it not to be relevant.
Also interesting:
- episode 1 mention of Mrs. Houseman talks about her "friends from New York" and how they "keep the spa" running.
- James went down to the boiler room and saw that creepy door.
Boiler room, spa, weird humming, Mrs. Houseman.
Now if only her first name would be revealed as "Judith Houseman".
Now we've heard it both at The Great Northern and in the Las Vegas hospital. It seemed to be luring Bushnell out of there. So what possible reason could the humming sound have for The Great Northern?
I'm wondering if the humming in the Great Northern and the hospital are happening concurrently - i haven't pieced together all the scrambled timelines, has anybody else and is this possible?

I'm a big fan of the "Scrambled timeline theory".
If the humming is indeed a clue it really could be a big help in putting everything in the right chronological order. It could be very frustrating though. If the finale will be messed up (in a good way) as I think it will be, we will have plenty of time to patch everything together after next week!

It seems, if nothing else, to elicit a desire on people's parts to follow it to find its source. Maybe that is its purpose! It led James to that door. Sure wish I know what he saw there.
Whatever it was, James didn't seem bothered as he went to the Roadhouse with Freddie. If the humming was due to Mike, Laura or something else, it would have rattled him and he would have called out his superhero friend. Maybe he got scared and didn't open the door.

After Cooper's awakening, it seems clear that the hum has something to do with Lodge entities. Beverly and Ben weren't scared by it, so I don't see why James would be.
Doesn't The Arm make the same sound when he rubs his hands together in the original series? The sound is from a Tibetan singing bowl which fits with a number of themes in and influences on Twin Peaks.
